The analysis of cyclin D1 gene in squamous cell carcinoma of the larynx

Introduction: G870A polymorphism in 241 codon of the CCND1 gene has an influence on the frequency of alternative splicing and formation of altered protein. The changed cyclin D1 can have a longer half-life time. CyclinD1 plays a key role in the regulation of cell cycle, and its over-expression is found in the squamous cell carcinoma of the larynx. G/A polymorphism in CCND1 gene could be a genetic factor modifying the individual susceptibility to laryngeal cancer. Material and Methods: The G/A genotype frequency and alleles distribution in 63 patients with laryngeal cancer and within the control group of 102 healthy patients were analysed. In cancer patients the immunohistochemical evaluation was performed. The DNA was extracted from the parrafin embadded tissue in the cancer patients, and from the peripheral blood within the control group. The genotypes were analyzed with use of the PCR-RFLP method. The statistical analysis (OR, 95% Confidence intervals (CI) and p) was performed using the GraphPad Prism v.3.0. Results: GA genotype was related to increased cancer risk (OR-3,02 95% CI-1, 39-6,56; p=0,0045) The frequency of A allele (0,484) was higher in cancer patients than in healthy ones(0,416) and was associated with slight increase of laryngeal cancer risk (OR-1,34 95% CI-0,84-2,05) Homozygotic AA genotype correlates with the presence of metastases in the lymph nodes. The correlation between the level of cyclinD1 staining in IHC study and AA genotype has been found. Conclusions: The results of our studies suggest that the polymorphism of the CCND1 gene may increase the individual susceptibility to the development of laryngeal cancer because: 1/ The homozygotic AA genotype correlates with the over-expression of cyclin D1 protein. 2/ The homozygotic AA genotype make the mean age of the onset lower. 3/ The homozygotic AA genotype significantly correlates with the presence of metastases to the lymph nodes.
